摘要: 目的:观察雷公藤甲素对人卵巢癌细胞株SKOV3增殖的抑制作用,与顺铂单独作用相比较,评价其抗卵巢癌活性。 方法:MTT法检测不同的用药方式对SKOV3细胞增殖的抑制作用;倒置显微镜观察雷公藤甲素作用后细胞的形态学变化;流式细胞仪检测雷公藤甲素对SKOV3细胞诱导凋亡情况。 结果:雷公藤甲素对SKOV3细胞增殖有显著的抑制作用,并呈量-效、时-效关系,24、48、72h的IC50分别为70.3、40.0、31.7 nmol/L。与顺铂单独作用相比较,其抑制率高于顺铂(24、48、72h的IC50分别为33.0、19.2、9.6 μmol/L)。倒置显微镜观察雷公藤甲素作用SKOV3后细胞生长明显受到抑制。流式细胞仪检测雷公藤甲素作用SKOV3后发生细胞凋亡。 结论:雷公藤甲素可显著抑制人卵巢癌细胞株SKOV3的增殖,并且其抑制率显著高于顺铂,其抗卵巢癌的作用可能是通过诱导细胞凋亡实现的。

Abstract: Objective: To evaluate the activity of triptolide as an anti-ovarian cancer drug by investigating the inhibitive effect of triptolide and comparing with cisplatin on human ovarian cancer cell line SKOV3. Methods: SKOV3 cells were treated with different concentrations of triptolide and cisplatin respectively, the anti-cancer activity was evaluated by MTT assay. The cells' morphologic change affected by triptolide was observed by microscope. And the cells' apoptosis treated by triptolide was detected by FCM. Results: MTT assay showed that triptolide could significantly inhibit the proliferation of SKOV3 cells in a dose-dependent and time-dependent manner, and the half maximal inhibitory concentration (IC50) of 24, 48, 72h were 70.3、40.0、31.7 nmol/L. Comparing with cisplatin, triptolide had more significant inhibitive effect on SKOV3 cells, for the same time, the IC50 of cisplatin were 33.0、19.2、9.6 μmol/L. Inverted microscope observed that cells growth were obviously inhibited. Apoptosis existed in SKOV3 cells after treatment with triptolide. Conclusion: Triptolide has significant suppressive effect on SKOV3 cells, and the inhibitive rate of triptolide was higher than that of cisplatin. The anti-tumor effect of triptolide may be achieved by inducing cell apoptosis.
